The nonkeratinized stratified squamous epithelium of the vagina and ectocervix consists of four distinct histological layers from deep to superficial: 1) Stratum basale (basal cell layer), 2) Stratum parabasale (parabasal cell layer), 3) Stratum spinosum (intermediate cell layer), and 4) Stratum superficiale (superficial cell layer). The stratum corneum is the outermost layer of keratinized epithelium found in skin, characterized by dead, anucleated cells filled with keratin, but this layer is absent in the nonkeratinized mucosal epithelium of the female genital tract. The absence of keratinization allows the vaginal and cervical epithelium to remain moist and flexible, which is essential for its protective and functional roles.
