4E-BP1 (eIF4E-binding protein 1) is a key inhibitor of cap-dependent translation initiation. In its hypophosphorylated state, it binds tightly to the translation initiation factor eIF4E, preventing it from assembling the eIF4F complex needed to start protein synthesis. mTORC1 phosphorylates 4E-BP1, causing it to release eIF4E. This freed eIF4E can then participate in translation, leading to increased synthesis of proteins that promote cell growth and proliferation, many of which are involved in cancer.
