Many environmental toxicants function as anti-androgens by acting as androgen receptor antagonists. This means the chemical, such as the fungicide vinclozolin or the DDT metabolite p,p'-DDE, has a molecular shape that allows it to bind to the androgen receptor—the same receptor that natural testosterone uses. However, when the antagonist binds, it does not activate the receptor. Instead, it physically blocks the binding site, preventing the natural testosterone from attaching and sending its normal signal. It's like putting the wrong key in a lock; it jams the lock so the right key can't turn it. This blockade can lead to 'demasculinizing' effects during development, such as reduced size of male reproductive organs, hypospadias, and impaired sperm production later in life.
