Mucolytic agents are essential for processing sputum samples because they break down the thick, viscous mucus that can obscure cellular material during microscopic examination. Commonly used agents include 8% hydrochloric acid, which effectively digests mucin, and commercial preparations based on cysteine derivatives, such as Cytoclair. These agents work by disrupting disulfide bonds in glycoproteins, liquefying the mucus and releasing trapped cells.
