The MCV Small T (sT) antigen is a potent oncoprotein. A key mechanism is its binding to and inhibition of the cellular protein phosphatase 2A (PP2A). PP2A is a major tumor suppressor that dephosphorylates and regulates many signaling proteins involved in growth and survival. By inhibiting PP2A, sT leads to hyperphosphorylation and dysregulation of its client proteins, including those in the mTOR and cap-dependent translation pathways. This drives increased protein synthesis, cell growth, and survival, contributing to the transformation of Merkel cells.
