While the Column Technique (also known as gel technology) is a modern and widely used method in blood banks, it is an automated or semi-automated system, not a classic manual technique. Traditional manual methods for ABO grouping include the tube technique, the slide or tile method (using a milk glass plate), and card methods. These manual techniques rely on visual assessment of agglutination in a liquid or on a solid surface. The column technique uses a gel matrix in microtubes to trap agglutinated cells, and while it can be performed manually, it represents a more advanced technological platform compared to the basic tube or slide methods.
