Q: What type of tissue preparation is most suitable for enzyme histochemistry?

This statement is true. The Indirect Antiglobulin Test (IAT), also called the indirect Coombs test, detects antibodies that are freely circulating in a person's serum or plasma. In this test, the patient's serum is incubated with commercially prepared red blood cells of known antigen type. If the serum contains antibodies against antigens on these test cells, the antibodies will bind. The cells are then washed and anti-human globulin (Coombs' reagent) is added. If antibodies have coated the cells, agglutination occurs—a positive IAT. This test is primarily used for: 1) Antibody screening and identification in pre-transfusion testing, and 2) Detecting anti-D and other antibodies in prenatal screening of pregnant women. It is different from the Direct Antiglobulin Test (DAT), which detects antibodies already attached to the patient's own red cells in vivo.
