The Modified Ziehl-Neelsen stain (or Kinyoun stain) is an acid-fast stain used primarily for detecting certain bacteria (like Mycobacterium tuberculosis) and certain coccidian parasites in stool, specifically the oocysts of Cryptosporidium parvum, Cyclospora cayetanensis, and Cystoisospora belli. These oocysts retain the red carbol fuchsin dye after acid-alcohol decolorization. It is not used for diagnosing intestinal amoebiasis caused by Entamoeba histolytica. The gold standard for diagnosing amoebiasis is the microscopic identification of E. histolytica trophozoites or cysts in stool samples, often using permanent stains like Trichrome or Iron Hematoxylin that allow for visualization of nuclear detail. Antigen detection tests and PCR are also used. Confusing these staining techniques can lead to misdiagnosis.
