KSHV encodes v-Cyclin, a viral homolog of cellular cyclin D. v-Cyclin binds to and activates cellular CDK6, promoting phosphorylation and inactivation of the retinoblastoma (pRb) tumor suppressor protein. This pushes the infected cell from G1 into S-phase of the cell cycle, contributing to uncontrolled proliferation seen in KSHV-associated cancers like Kaposi's sarcoma.
