The ABC method is an indirect detection technique that provides significant signal amplification, making it much more sensitive than the direct method. In the ABC method, after the primary antibody binds, a biotinylated secondary antibody is applied. This secondary antibody recognizes the primary antibody. Then, a preformed complex of avidin and biotinylated enzyme (the ABC complex) is added. Avidin has an extremely high affinity for biotin, so the complex binds tightly to the many biotin molecules on the secondary antibody. Importantly, the ABC complex itself contains multiple enzyme molecules. This multi-layered approach means that for each primary antibody molecule, many enzyme molecules are ultimately localized to the site, creating a powerful amplified signal. In contrast, the direct method conjugates the enzyme directly to the primary antibody, providing only one enzyme per primary antibody and thus much lower sensitivity.
