True. The LSAB (Labeled StreptAvidin-Biotin) method is a classic and highly sensitive three-step IHC detection technique. The steps are: 1) Application of the unlabeled primary antibody. 2) Application of a biotinylated secondary antibody that recognizes the primary antibody. 3) Application of enzyme-conjugated streptavidin (e.g., streptavidin-horseradish peroxidase). The streptavidin, with its extremely high affinity for biotin, binds to the biotin tags on the secondary antibody, localizing the enzyme to the antigen site. This method provides excellent signal amplification because multiple biotin molecules are on the secondary antibody and each streptavidin conjugate can carry multiple enzyme molecules. It was a significant improvement over the ABC method because streptavidin's near-neutral charge reduces non-specific ionic binding to tissue compared to positively charged avidin, resulting in lower background. While still used, it has been largely supplanted in many labs by even simpler two-step polymer-based systems.
