The presentation illustrates several methods for performing ABO blood grouping. Two classic and commonly used manual techniques are the Tube Technique (Röhrchentechnik) and the Slide or Milk Glass Plate method. In the Tube Technique, patient red cells or serum are mixed with the appropriate reagents (antisera or reagent cells) in test tubes, centrifuged, and then observed for agglutination. This method is sensitive and allows for grading the strength of reactions. The Slide or Milk Glass Plate method involves placing a drop of patient blood and a drop of antiserum on a clean glass plate or tile, mixing them, and gently rocking the plate while observing for agglutination. This method is rapid and simple but is generally less sensitive than the tube method and is often used for preliminary screening or in resource-limited settings.
