In a direct IHC method, the key component labeled with the detectable tag (such as an enzyme or fluorochrome) is the primary antibody itself. This means the antibody that specifically recognizes the target antigen is directly conjugated to the label. The procedure is therefore a single-step incubation: the labeled primary antibody is applied to the tissue, it binds directly to its antigen, and the location is visualized. This method is simple and fast, with low risk of cross-reactivity because no secondary antibody is used. However, it lacks signal amplification, as only one label molecule is delivered per primary antibody. This makes it less sensitive than indirect methods and is why its use is largely confined to applications where the antigen is abundant, such as detecting immunoglobulins in renal biopsies by direct immunofluorescence.
