Insulin is a large protein hormone. It cannot cross cell membranes by simple diffusion or through channels. When injected subcutaneously, it forms a depot in the tissue fluid. From there, it enters the bloodstream primarily through the lymphatic system and capillaries. The movement from the injection site into these vessels occurs via bulk flow through pores and spaces between cells in the capillary walls, a process often referred to as convective transport or paracellular absorption.
