The Prussian blue stain is the standard cytochemical method for detecting non-heme (storage) iron in tissues and cells. In the bone marrow, it reacts with ferric iron (Fe3+) in hemosiderin within macrophages to produce a bright blue color. It is also used to see if developing red blood cell precursors (erythroblasts) have abnormal iron-loaded mitochondria circling the nucleus, which are called ring sideroblasts—a key finding in certain types of anemia like sideroblastic anemia.
