SOCS1 (Suppressor of Cytokine Signaling 1) is a critical member of the SOCS family that provides negative feedback. Upon STAT-induced gene expression, SOCS1 is produced. It inhibits JAK-STAT signaling through multiple mechanisms: 1) It binds to and directly inhibits JAK kinase activity. 2) It binds to phosphorylated tyrosine residues on cytokine receptors, blocking STAT recruitment. 3) It acts as an E3 ubiquitin ligase adaptor, targeting bound proteins (like JAKs and receptors) for proteasomal degradation. This tightly controls the duration and intensity of cytokine signals.
