AKT, when activated by the PI3K pathway, phosphorylates TSC2 (a component of the complex). This phosphorylation inhibits the GAP activity of the TSC1/TSC2 complex. As a result, Rheb accumulates in its active GTP-bound form. Active Rheb-GTP directly binds to and activates mTORC1, stimulating protein synthesis and cell growth.
