In the absence of a WNT signal, the 'destruction complex' (Axin, APC, GSK3β, CK1) captures β-Catenin. It sequentially phosphorylates β-Catenin, which acts as a signal for its ubiquitination and subsequent degradation by the proteasome. This keeps cytoplasmic β-Catenin levels low and prevents inappropriate gene activation.
