Rheb (Ras homolog enriched in brain) is a small GTPase that is a direct and essential activator of mTORC1. When Rheb is in its active, GTP-bound state (a condition promoted by the inactivation of the TSC1/TSC2 complex by AKT), it binds to and allosterically activates the kinase activity of mTOR within the mTORC1 complex, stimulating protein synthesis and cell growth.
