APC (Adenomatous Polyposis Coli) is the 'gatekeeper' tumor suppressor gene in colorectal cancer. It is mutated in approximately 80% of sporadic colorectal cancers and in Familial Adenomatous Polyposis (FAP). The APC protein serves as the central scaffold of the destruction complex, binding to Axin, β-catenin, and CK1/GSK3β. This facilitates the efficient phosphorylation and degradation of β-catenin. Loss of APC function leads to β-catenin accumulation and constitutive WNT pathway activation.
