Saturation kinetics (zero-order kinetics) occurs when the enzymes responsible for metabolizing a drug become fully occupied or saturated. At this point, the rate of metabolism becomes constant and does not increase even if more drug is present. This is important because small increases in dose can lead to large, unpredictable increases in drug concentration in the blood, dramatically raising the risk of toxicity. Drugs like phenytoin and alcohol exhibit this behavior, requiring careful dosing.
