The main advantage of using a tissue microarray (TMA) in IHC validation or research is the ability to analyze a large number of different tissue samples simultaneously under identical staining conditions. A TMA is constructed by extracting small cylindrical cores (typically 0.6-2.0 mm in diameter) from many donor paraffin blocks (e.g., hundreds of different tumors, normal tissues, or cell line pellets) and embedding them in a single recipient paraffin block. A single section from this block contains all these mini-tissue samples. When this TMA slide is stained with a specific antibody, the researcher gets a snapshot of that marker's expression across dozens to hundreds of cases on one slide. This enables high-throughput, consistent, and comparative analysis. It maximizes resource efficiency (using minimal reagent), reduces inter-slide staining variability, and allows for rapid validation of antibody specificity and assessment of biomarker prevalence across different disease types or stages.
