The Harada-Mori technique is a simple, low-cost culture method specifically designed for the diagnosis of intestinal nematodes (roundworms) that hatch larvae in stool, primarily Strongyloides stercoralis and hookworm species (Ancylostoma duodenale and Necator americanus). A small amount of fresh stool is placed on a strip of filter paper, which is then partially immersed in water in a tube. The tube is sealed and kept at room temperature for several days. Larvae present in the stool (or that hatch from eggs) migrate down the moist filter paper into the water. The water is then examined microscopically for the presence of characteristic larvae. This technique concentrates and allows for the identification of larvae based on their morphology (e.g., the buccal cavity and genital primordium of Strongyloides vs. hookworm). It is not used for protozoa like Giardia or Entamoeba.
