Pharmacodynamic tolerance refers to adaptive changes in the body that reduce its response to a drug, even if the drug concentration at the site of action remains the same. A common mechanism for opioid tolerance is receptor down-regulation—a decrease in the number of active opioid receptors on the surface of neurons. With fewer receptors available, the same dose of morphine produces a weaker effect.
