The GLI family of zinc-finger transcription factors (GLI1, GLI2, GLI3) are the final mediators of Hedgehog signaling. In the canonical pathway, activation of SMO leads to the conversion of full-length GLI proteins into their transcriptional activator forms, which then enter the nucleus and regulate the expression of target genes involved in cell proliferation and stemness.
