The biotin-streptavidin (B-SA) method offers a significant advantage over the older avidin-biotin complex (ABC) method by using **streptavidin** instead of avidin. Avidin, a protein from egg whites, is positively charged and contains sugar (carbohydrate) groups. These properties can cause it to bind non-specifically to negatively charged structures and lectin-like molecules in tissues (like in kidney, liver, and brain), leading to high background staining or false positives. **Streptavidin**, derived from a bacterium, has a near-identical affinity for biotin but is neutrally charged and contains no carbohydrates. This means it has much lower non-specific binding to tissue, resulting in a cleaner background and more specific staining. Furthermore, streptavidin-enzyme conjugates are highly stable reagents, allowing them to be prepared in ready-to-use forms and stored for long periods, improving consistency and ease of use in the diagnostic laboratory.
