PTEN is a lipid phosphatase. Its primary and most critical tumor-suppressive function is to dephosphorylate phosphatidylinositol (3,4,5)-trisphosphate (PIP3) at the 3-position, converting it back to phosphatidylinositol (4,5)-bisphosphate (PIP2). This action directly antagonizes PI3K and shuts off the pro-growth PI3K/AKT/mTOR signaling cascade.
