PTCH1 acts as a receptor and inhibitor for the Hedgehog pathway. In the absence of the Hedgehog signal, PTCH1 suppresses the activity of the SMO protein. Mutations in PTCH1, which inactivate it, are a common cause of constitutive pathway activation in cancers like basal cell carcinoma.
