A person with blood type AB has both A and B antigens on their red blood cells. In forward grouping (cell typing), the patient's red cells are mixed separately with commercial antisera. Anti-A serum contains antibodies that specifically bind to A antigens. Anti-B serum contains antibodies that specifically bind to B antigens. Therefore, the red cells from a type AB individual will agglutinate (clump) when mixed with both anti-A and anti-B reagents. This pattern of agglutination with both antisera is diagnostic for blood type AB. It is the opposite of type O, which shows no agglutination with either reagent.
