The HIF (Hypoxia-Inducible Factor) pathway is the primary cellular response to low oxygen levels (hypoxia). In normoxia, HIF-α subunits are hydroxylated, recognized by the VHL E3 ubiquitin ligase, and degraded. In hypoxia, this degradation is blocked, allowing HIF-α to accumulate, dimerize with HIF-β, and translocate to the nucleus. There, it activates the transcription of hundreds of genes involved in adaptation to hypoxia, including VEGF (vascular endothelial growth factor), which stimulates the growth of new blood vessels (angiogenesis) to supply the tumor with oxygen and nutrients.
