Studying a hematoxylin and eosin (H&E) stained section from the donor block is an essential pre-analytical, quality control step known as 'pathologist review' or 'block annotation.' This step is not about processing the tissue; it is about intellectual triage and precise planning. The H&E slide provides a detailed two-dimensional map of the three-dimensional tissue in the block. A pathologist or trained researcher examines this slide to identify and mark the exact morphological region of interest (ROI). For example, in a tumor block, they need to distinguish viable tumor from areas of necrosis, fibrosis, normal tissue, or inflammation. The goal is to ensure that the tiny core taken for the TMA is representative of the biological question. By marking the specific ROI on the H&E slide (or a corresponding digital image), they create a guide that ensures the tissue microarray instrument's punch targets the correct spot, guaranteeing that the resulting data is scientifically valid and relevant.
