Mucolytic agents such as 8% hydrochloric acid are used to break down the thick, viscous mucus present in sputum samples. This mucus can trap cellular elements and obscure them during microscopic examination, making it difficult to identify abnormal cells or pathogens. By digesting the mucin, these agents release the trapped cells and allow for better concentration during centrifugation. The resulting cellular pellet yields clearer and more diagnostic smears, improving the detection of malignant cells, infectious organisms like fungi or acid-fast bacilli, and other diagnostic features.
