Affinity refers to the strength of binding between a drug and its receptor. A drug with high affinity binds tightly and readily to the receptor, even at low concentrations. Loratadine is an antagonist—it binds to the H1 receptor but does not activate it. Its effectiveness depends largely on its ability to bind tightly (high affinity) and block histamine from binding.
