It is standard practice to submit three consecutive early morning sputum specimens for cytological examination. This approach increases the likelihood of obtaining a representative sample from the lower respiratory tract and improves diagnostic sensitivity. Early morning samples are preferred because they are more likely to contain material that has accumulated in the lungs overnight, providing a richer cellular yield. Collecting multiple samples reduces the chance of false-negative results due to sampling error or contamination from saliva or upper respiratory secretions.
