Rapamycin (also known as sirolimus) is a macrolide compound originally discovered as an antifungal agent. It binds to the intracellular protein FKBP12, and this complex potently and specifically inhibits mTORC1 (but not mTORC2). Due to its immunosuppressive effects on T-cell proliferation, it is used to prevent organ transplant rejection. Its ability to inhibit the central growth regulator mTORC1 also gives it anti-cancer properties, leading to the development of rapamycin analogs (rapalogs) like everolimus and temsirolimus for cancer treatment.
