The TSC1/TSC2 complex acts as a GTPase-activating protein (GAP) for the small G-protein Rheb. In its active state, it keeps Rheb in its inactive GDP-bound form. When AKT phosphorylates and inhibits TSC2, Rheb is allowed to switch to its active GTP-bound state. Active Rheb-GTP directly activates mTORC1, the complex that stimulates protein synthesis, cell growth, and metabolism.
