c-Myc is a key proto-oncogene and transcription factor that drives cell cycle progression. One of the primary mechanisms by which TGF-β induces G1 arrest is through the repression of c-Myc expression. The SMAD complexes, in cooperation with co-repressors like E2F4/5 and p107, bind to the c-Myc promoter and actively repress its transcription. Lower levels of c-Myc protein lead to decreased expression of genes required for G1/S transition. In cancers resistant to TGF-β growth inhibition, c-Myc repression often fails.
