PTEN's main function is to dephosphorylate PIP3 back to PIP2, turning off the PI3K signal. Loss of PTEN removes this critical 'off switch.' As a result, PIP3 accumulates, leading to persistent and unregulated activation of the entire downstream PI3K/AKT/mTOR pathway, which drives cell growth, survival, and metabolism—hallmarks of cancer.
