Naive T cell activation requires two signals. Signal 1 is antigen-specific: the TCR binding to peptide-MHC on the APC. Signal 2 is a co-stimulatory signal, most crucially the binding of B7 molecules (CD80/CD86) on the APC to CD28 on the T cell. Without Signal 2, the T cell may become anergic (unresponsive).
