Permanent stains, such as Trichrome or Iron Hematoxylin, are applied to fixed stool smears and create a lasting slide. Their primary advantage is the superior morphological detail they provide. They stain internal structures—nuclei, karyosomes, chromatoid bodies, axonemes, and flagella—with high contrast and clarity. This allows for precise identification and differentiation of protozoan trophozoites and cysts based on subtle nuclear and cytoplasmic features, which is essential for distinguishing pathogenic from non-pathogenic species (e.g., Entamoeba histolytica from Entamoeba coli). Temporary wet mounts (saline or iodine) are quick and useful for observing motility and general shape, but they do not provide the same level of cytological detail, and the preparation dries out quickly. Permanent stains are indispensable for definitive diagnosis and for maintaining archival records.
