In an Indirect ELISA, the solid phase (well) is coated with HIV antigen. If the patient's serum contains HIV antibodies, they bind to the antigen. An enzyme-linked anti-human antibody (conjugate) is then added, which binds to the patient's antibodies. A substrate is added, and the enzyme causes a color change, indicating a positive result.
