Rapamycin (also called sirolimus) is an mTOR inhibitor. It binds to FKBP12 and this complex then inhibits mTOR, a key protein in cell growth and proliferation signaling pathways. By blocking mTOR, rapamycin prevents T cells from proliferating in response to IL-2 and other growth signals, suppressing the immune response against transplants.
