p27KIP1 (and its related protein p21CIP1) are members of the CIP/KIP family of CDK inhibitors. They bind to and inhibit the activity of Cyclin-CDK complexes, particularly Cyclin E-CDK2 and Cyclin D-CDK4/6, acting as a brake on the G1/S transition. Their degradation or downregulation, often mediated by oncogenic pathways, is a common event in cancer, allowing unchecked cell cycle progression.
