Most Gastrointestinal Stromal Tumors (GISTs) are driven by activating mutations in the KIT gene, which encodes a receptor tyrosine kinase (RTK). These mutations lead to ligand-independent dimerization and constitutive activation of the KIT receptor. This, in turn, primarily activates the PI3K/AKT/mTOR and the MAPK (RAS/RAF/MEK/ERK) signaling pathways, promoting cell survival and proliferation. This understanding led to the successful use of the tyrosine kinase inhibitor imatinib as a targeted therapy for GIST.
