The LKB1-AMPK axis acts as a key energy-sensing brake on mTORC1. Under conditions of low cellular energy (high AMP/ATP ratio), AMPK is activated by LKB1. Activated AMPK then phosphorylates and activates the TSC1/TSC2 complex, enhancing its GAP activity towards Rheb. This leads to Rheb inactivation, which directly inhibits mTORC1. Additionally, AMPK can phosphorylate Raptor, a component of mTORC1, further inhibiting its activity. This conserves energy by shutting down anabolic processes like protein synthesis when energy is scarce.
